Establishment of an acute rejection model of rat orthotopic liver transplantation

چکیده
BACKGROUND: Some research indicates that the Wistar and Sprague Dawley rats belong to closed colony strains, rather than inbred strains, which not only has some hereditary stability but also genetic polymorphism. Thus, the Wistar to SD rat liver transplantation models may not be the ideal models to study the acute rejection of rat liver transplantation. OBJECTIVE: To establish an acute rejection model of Lewis-BN rat orthotopic liver transplantation. METHODS: The model was established by modified Kamada’s two-cuff technique. The rats were performed Lewis to Lewis or Lewis to BN rat liver transplantation. The pathological changes of transplanted livers were observed and levels of alaine aminotransferase (ALT) and total bilirubin (TBIL) were detected at 3, 5 and 7 days after transplantation. The survival time of rats was determined. RESULTS AND CONCLUSION: There was no rejection in isogene group from Lewis to Lewis rat, and the average survival time was over 100 days. Liver function was mildly impaired. Conversely, the average survival time was (12.75±1.25) days in the allogene group. The pathological examination showed a typical acute rejection at 7 days. Liver function was severely impaired. At the same time point after the operation, levels of ALT and TBIL in the allogene group were significantly higher than those of the isogene group (P < 0.05). The results indicate that, Lewis to BN rat combination is a stable acute rejection model of rat liver transplantation, but there may be technical difficulties due to its poor tolerance. The sophisticated microsurgical technique and the delicate surgical manipulation is the key to achieve success.

Severe Acute Hyperkalemia during Pre-Anhepatic Stage in Cadaveric Orthotopic Liver Transplantation
A serious hazard to patients during orthotopic liver transplantation is hyperkalemia. Although the most frequent and hazardous hyperkalemia occurs immediately after reperfusion of the newly transplanted liver, morbid hyperkalemia could happen in the other phases during orthotopic liver transplantation. However, pre-anhepatic hyperkalemia during orthotopic liver transplantation is rare. This rep...
متن کاملOrthotopic Liver Transplantation for Etanercept-Induced Acute Hepatic Failure; a case report
The occurrence of hepatotoxicity following etanercept (tumor necrosis factor-alpha antagonist) prescription, has been studied well. However, an acute hepatic failure leading to liver transplant as an adverse effect of this drug has not been reported in the literature. In this article, we are going to present a case of acute liver failure followed by liver transplantation, in a 32 years old man ...
متن کاملExperimental and clinical significance of antinuclear antibodies in liver transplantation.
Histone H1 and high-mobility group box 1 (HMGB1) proteins are known to initiate an immune reaction, and the corresponding antibodies (Abs) possess immunosuppressive activity. In the present study, we aimed to evaluate the immunological role of antinuclear Abs in experimental and clinical liver transplantation. In a rat tolerogenic orthotopic liver transplantation (OLT) model, antihistone H1 and...

The mice in the experimental group were divided into three groups according to the warm ischemia time (WIT) of liver graft: normal LTx, WIT 30 minut...
متن کاملExpression of vascular endothelial growth factor and basic fibroblast growth factor in acute rejection reaction following rat orthotopic liver transplantation
The aim of the present study was to investigate the expression levels of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) in acute rejection reaction (ARR) following orthotopic liver transplantation in a rat model. Serum VEGF and bFGF levels were detected using ELISA, and their expression levels in liver and spleen tissues were determined using immunohistochem...
